Scrub typhus, also known as scrub typhus, is an important zoonotic disease prevalent in Asia and the Pacific. The pathogen causing the disease is *Orientia tsutsugamuchi*, an obligate intracellular parasite, Gram-negative, with several antigen-specific serotypes, including Gilliam, Karp, Kato, and Kawasaki. In my country, scrub typhus is widespread. Wild rodents are the natural reservoir hosts, and humans are primarily infected through bites from the chigger mite during outdoor activities. Because it lacks characteristic clinical symptoms (primarily fever, with some patients exhibiting eschar), patients are often misdiagnosed or missed, leading to serious consequences.

The 56 kDa recombinant antigen was expressed using an *E. coli* expression system and purified by Ni affinity chromatography, achieving a purity of over 95% (Fig. 1).
