Ovalbumin (OVA) is the main protein component of egg white, accounting for 54%-69% of the total protein content. OVA is a typical globulin with a molecular weight of 44.5 kDa, belonging to the phosphoglycoprotein family, containing four free sulfhydryl groups and 385 amino acid residues. OVA is a major allergen causing egg allergies in humans. Some viral vaccines, such as influenza and yellow fever vaccines, use chicken embryos as a culture medium; therefore, monitoring the residual OVA content in these vaccines has become an essential testing item in the production process.
In this project, we prepared monoclonal antibodies by immunizing mice with natural and purified OVA. We used a double-antibody sandwich ELISA platform to screen for paired antibodies, which showed a linear relationship within the range of 8-64 ng/ml (Fig. 1). No cross-reactivity was observed with bovine serum, human serum albumin, rabbit serum, or horse serum, indicating good specificity.
A double-antibody sandwich ELISA kit for detecting OVA was constructed using this paired monoclonal antibody. Evaluation showed that the kit exhibits good stability (6 months to 1 year) and low batch-to-batch variability (CV ≤ 15%), making it suitable for manufacturers to detect OVA residues during vaccine production.
