Scrub typhus, also known as scrub typhus, is an important zoonotic disease prevalent in Asia and the Pacific. The pathogen causing the disease is *Orientia tsutsugamuchi*, an obligate intracellular parasite, Gram-negative, with several antigen-specific serotypes, including Gilliam, Karp, Kato, and Kawasaki. In my country, scrub typhus is widespread, with wild rodents serving as natural reservoir hosts. Humans are primarily infected through bites from the chigger mite during outdoor activities. Due to the lack of characteristic clinical symptoms (primarily fever, with some patients exhibiting eschar), patients are often misdiagnosed or missed, leading to serious consequences. Our company has developed a human IgM antibody diagnostic kit targeting the 56 kDa antigen of the pathogen, for both early diagnosis of human infection and confirmation of past infections. The total antibody diagnostic kit can also be used for seroepidemiological surveys in humans and animals. Validated on a large number of clinical specimens, this kit demonstrates good specificity and sensitivity, effectively identifying serotypes such as Gilliam, Karp, Kato, and Kawasaki.
Using anti-human μ-chain monoclonal antibody as the capture antibody and HRP-labeled 56 kDa protein as the diagnostic antigen, a capture ELISA detection system (IgM antibody capture ELISA, MAC-ELISA) is constructed. This system detects Orientia scrub typhus-specific IgM antibodies in the serum of infected individuals during the acute phase, enabling early diagnosis.
