Numerous studies have shown that the VP30 (viral protein 30) protein encoded by the ASFV genome possesses pathogen specificity and is one of the candidate target molecules for laboratory diagnostics. Nanjing Miaodi Biotechnology Co., Ltd. has developed a recombinant VP30 antigen and its paired monoclonal antibody, which is provided as raw materials to immunodiagnostic reagent companies. This recombinant antigen uses the gene sequence of domestic ASFV isolates as a template, and is solublely expressed and purified in the *E. coli* system using gene recombination technology (Figure 1). Preliminary experimental results show that this protein exhibits good specificity and sensitivity in immunodiagnostic reagents such as ELISA and colloidal gold assays.
Mouses were immunized with this protein, and multiple murine monoclonal antibodies were prepared using classic hybridoma technology. Pairing experiments yielded antibody pairs with high specificity and sensitivity, which can be used in the development of ELISA and colloidal gold assay reagents. Experimental results show that these antibodies do not cross-react with classic classical swine fever and porcine infectious diarrhea pathogens, and the detection sensitivity in a double-antibody sandwich ELISA can reach 0.5-1.0 ng/ml (Figure 2).
